Biological Nanostructures Laboratory - Genetics Core

The Biological NanoStructures Lab (BNL) is a 1600 square foot shared multi-user environment for biological synthesis and analysis. Instrumentation for molecular biology, recombinant protein expression, purification & analysis and tissue culture will be available on a recharge basis.

This core provides Next Generation Sequencing (NGS) library prep and sequencing resources to on- and off-campus researchers near Santa Barbara, CA. NGS (aka High Throughput Sequencing) experiments are conducted on the Illumina NextSeq 500 which uses the same widely accepted SBS technology found in other popular Illumina sequencers such as the HiSeq and MiSeq.

Access Procedure:

Trained users will have keycard access to the lab and will be able to book and use the analytical instruments. For ease of management, molecular biology instruments located in the BNL are also part of the Analytical core. Researchers can also engage BNL staff to perform basic molecular biological experiments such as recombinant protein expression. Cost will be on a project-by-project basis and will include staff man-hours, materials used and instrument recharges.

Steps to Use Equipment
  1. Contact BNL staff to get trained on the instrument.
  2. Apply to CNSI for electronic door access to the facility.
  3. BNL Staff will add you to the FBS reservation system.

Instruments

  1. Agilent TapeStation

The TapeStation is used for nucleic acid analysis. Genomic DNA, RNA and prepped dsDNA libraries can be analyzed with the instrument to get size distribution and concentration estimates.

  2. Bio Rad QX200 Droplet Digital PCR

Precise and sensitive digital PCR for a wide variety of applications, flexible PCR chemistry with user-friendly software for experimental set up and analysis.

  3. Bio-Rad CFX96 Touch RT-PCR

The CFX96 Touch System is a high precision, flexible Real-Time quantitative PCR system. The CFX96 Touch combines advanced optical technology with precise temperature control, for a fast and sensitive detection system for singleplex and multiplex reactions.

  4. Biomek 4000 Automated Liquid Handler

The Biomek 4000 Automated Workstation is a liquid handler for robotic pipetting of 1-1000 ul.

  5. Covaris M220 Focused-Ultrasonicator

The Covaris M220 is designed for Next-Gen Sequencing applications requiring fragment sizes between 150bp and 5kb.

  6. Illumina MiSeq

The MiSeq desktop sequencing system allows access to focused applications such as metagenomics, targeted gene expression, and small genone sequencing, and more. Many output kits are available for up to 15 Gb of sequencing, 25 M reads, and 2 x 300 bp read lengths.

  7. Illumina NextSeq 500

The NextSeq 500 desktop sequencing system provides the flexible power and simplicity to analyze whole genomes, exomes, and transcriptomes.

  8. Nuaire Biological Safety Hood

Class II type A2 Biosafety Cabinet.

  9. Qubit 2.0 Fluorometer

The Qubit 2.0 Fluorometer is a benchtop fluorometer for the quantitation of DNA, RNA, and protein, using the highly sensitive and accurate fluorescence-based Qubit quantitation assays.

  10. Sage Science Blue Pippin and Pippin Prep

The Blue Pippin and Pippin Prep allow researchers to easily collect from a mixture of DNA fragment sizes, narrow fragment distributions as well as wide ranges of fragments with minimal effort. Maximum DNA fragment size 50 Kb. Up to 5 samples can be ran at once, with no possibility of cross-contamination.
